Although autophagy was initially considered a nonselective cellular process for bulk degradation, recent studies have highlighted how selective autophagy plays a crucial role in regulating cell biology. Autophagosome formation is dependent on the ATG8 family of proteins, which also play a key role in the recruitment of cargo into autophagic membranes. Although ATG8 is present as a single protein in yeast, in higher eukaryotes it has expanded to include several paralogs; LC3A, LC3B, LC3C, GABARAP, GABARAP-L1, and GABARAP-L2. This expansion is thought to provide increased flexibility and selectivity to autophagic responses. Among the LC3-family, LC3B is best studied and most widely used marker for autophagosomes, whereas LC3C is the member with the highest sequence divergence. By mapping the proximal interactome of ATG8-paralogs, LC3B and LC3C, we uncover a new LC3C-Endocytic-Associated-Pathway (LEAP) that recruits plasma membrane (PM) cargo, the Met RTK, and Transferrin R to autophagosomes. We provide mechanistic understanding of the LC3C interactome and LEAP dependent cargo degradation and how this may be altered and contribute to cancers.
