The research fof Julian Stingele has established and shaped a new area of genome stability. His work has resulted in the discovery of an entirely unanticipated DNA repair pathway, namely proteolytic DNA-protein crosslink repair by the SPRTN protease. His work has established defective DNA-protein crosslink repair as the molecular defect underlying Ruijs-Aalfs syndrome, which is caused by germline mutations in the SPRTN gene and results in premature ageing and early-onset hepatocellular carcinomas.
The Stingele group is working towards identifying the cellular processes causing the surprisingly high levels of endogenous DNA-protein crosslinks by (1) conducting genome-wide suppressor screens in cells engineered to express SPRTN patient variants and by (2) developing new methodologies to systematically identify crosslinked proteins by mass spectrometry. Moreover, his lab combines novel model substrates with structural biology to determine the molecular mechanism of the SPRTN protease and to develop small molecule SPRTN inhibitors. Stingele’s group employs a highly interdisciplinary strategy involving various approaches ranging from biochemistry, genetic engineering to functional assays in human cells and advanced imaging to address their basic cancer research ques-tions. His findings have so far been recognized in the form of a prestigious EMBO Young Investigator Award in 2020 as well as a Bayer Early Excellence in Science Award in 2019. In addition he has been awarded a highly competitive ERC Starting Grant (2019).
